Credit: The Wistar Institute PHILADELPHIA and MELBOURNE, Australia (May. 17, 2021) A team of scientists from The Wistar Institute in Philadelphia and the Peter MacCallum Cancer Center in Melbourne, Australia, discovered a new checkpoint mechanism that fine-tunes gene transcription. As reported in a study published in Cell, a component of the Integrator protein complex tethers the protein phosphatase 2A (PP2A) to the site of transcription allowing it to stop the activity of the RNA polymerase II enzyme (RNAPII). Disruption of this mechanism leads to unrestricted gene transcription and is implicated in cancer. The study points to new viable opportunities for therapeutic intervention demonstrating the anti-cancer effect of a new combination treatment in preclinical models of solid and hematopoietic malignancies. ....
E-Mail Credit: UEF/ Raija Törrönen Researchers at the University of Eastern Finland have discovered previously unknown non-coding RNAs (ncRNAs) involved in regulating the gene expression of vascular endothelial growth factors (VEGF), the master regulators of angiogenesis. The study, conducted by the research groups of Associate Professor Minna Kaikkonen-Määttä and Academy Professor Seppo Ylä-Herttuala, provides a better understanding of the complex interplay of ncRNAs with gene regulation, which might open up novel therapeutic approaches in the future. The results were published in the Molecular and Cellular Biology Journal. Over the past years, the development of next generation sequencing techniques has revealed that around 97% of the human transcriptome is transcribed as non-coding RNAs, and although the role of the vast majority remains uncharacterized, many functions such as gene regulation have been proven. ....

The compound, which targets the vascular adhesion protein 1 (VAP-1) that regulates inflammatory cell traffic, is the first radiopharmaceutical that has been developed completely in Finland and has advanced to clinical trials. In the study that started with healthy volunteers, the radiopharmaceutical was found to be well tolerated and safe. The radiopharmaceutical is 68Ga-labelled Siglec-9 peptide. ....
