Coinfection of Clonorchis sinensis and hepatitis B virus: clinical liver indices and interaction in hepatic cell models | Parasites & Vectors

In China, people infected with hepatitis B virus (HBV) are commonly found in areas with a high prevalence of Clonorchis sinensis, a trematode worm. Published studies have reported that the progression of hepatitis B is affected by coinfection C. sinensis. Clinical data from a total of 72 patients with C. sinensis and HBV (as sole infection or with coinfections) and 29 healthy individuals were analysed. We also incubated the hepatic stellate cell line LX-2 with total proteins from C. sinensis adult worms (CsTPs) and HBV-positive sera. In addition, the human hepatoblastoma cell line HepG2.2.15 was treated with the antiviral drug entecavir (ETV), CsTPs and the anti-C. sinensis drug praziquantel (PZQ). Our clinical data indicated that the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TB) and hyaluronic acid (HA) were significantly higher in patients with coinfection than in those infected with HBV only. In cell model ....

Ring stage classification of Babesia microti and Plasmodium falciparum using optical diffraction 3D tomographic technique | Parasites & Vectors

Babesia is an intraerythrocytic parasite often misdiagnosed as a malaria parasite, leading to inappropriate treatment of the disease especially in co-endemic areas. In recent years, optical diffraction tomography (ODT) has shown great potential in the field of pathogen detection by quantification of three-dimensional (3D) imaging tomograms. The 3D imaging of biological cells is crucial to investigate and provide valuable information about the mechanisms behind the pathophysiology of cells and tissues. The early ring stage of P. falciparum were obtained from stored stock of infected RBCs and of B. microti were obtained from infected patients during diagnosis. The ODT technique was applied to analyze and characterize detailed differences between P. falciparum and B. microti ring stage at the single cell level. Based on 3D quantitative information, accurate measurement was performed of morphological, biochemical, and biophysical parameters. Accurate measurements of morphological parameter ....

Increased Collagen Type V α2 (COL5A2) in Colorectal Cancer is As

In the current study, DDR1 has been identified as a cancer-associated receptor tyrosine kinase that is highly expressed in several malignancies relative to normal tissues.
29 As an important receptor, DDR1 can bind all types of collagens and could influence their signalling upon collagen stimulation.
30 It has been reported that the phosphorylation of DDR1 could activate the WNT/β-catenin and AKT/mTOR signaling to drive cancer cell metastasis and progression.
31,32 We observed that pY792 DDR1, the active form of DDR1, was upregulated by COL5A2 overexpression in HCT116 and SW620 cells (Figure 4D).
These observations indicate that
COL5A2 expression upregulates WNT/β-catenin and PI3K/mTOR signaling via binding DDR1 with phosphorylation and activation of this receptor, which might contribute to CRC tumorigenesis or progression. ....
