Strontium (Sr) isotopes measured in fossil remains have been a useful tool to assess the geographical origin and even migrations of humans and other animals. In particular, dental enamel generally represents the ideal material, as it is dense and less prone to diagenetic replacement of Sr post-burial. However, fossil teeth can often be precious artefacts and difficult to access for destructive analysis. Here, we assess whether measuring Sr isotopes in fossil dental calculus could be used at least as a rangefinder to determine the geographical origin of an individual. We measured trace element concentrations in modern calculus (from a local dental practice), and trace element concentrations and 87Sr/86Sr ratios in human fossil calculus, dentine, and enamel from specimens collected in York, UK. Isotopes in fossil tooth enamel provide robust tools for reconstructing food webs, which have been understudied in Australian megafauna. To delineate the isotopic composition of primary consumers and understand dietary behaviour at the base of the food web, we investigate calcium (Ca) and strontium (Sr) isotope compositions of Pleistocene marsupial herbivores from Wellington Caves and Bingara (New South Wales, Australia). Sr isotopes suggest small home ranges across giant and smaller marsupial herbivores. Ca isotopes in Pleistocene marsupial herbivores cover the same range as those in modern wombats and placental herbivores. Early forming teeth are depleted in heavy Ca isotopes compared to late-forming teeth of a given individual, suggesting a weaning signal. Distinct Ca compositions between taxa can be interpreted as dietary niches. Abstract Trophic structures, i.e., the diets, food chain positions, and feeding relationships of species in an ecological community, are a fundamental yet understudied avenue of investigation into Australian megafauna extinction. Calcium (Ca) isotope analysis has been developed as a tool to reconstruct trophic levels in vertebrate palaeobiology and palaeoecology. A baseline of modern marsupial Ca isotope signatures in a single trophic level and dietary niche is required to successfully apply this tool in order to reconstruct Australian faunal food webs. We present Ca isotope data from dental enamel of modern Tasmanian bare-nosed wombats (Vombatus ursinus tasmaniensis) to constrain the Ca isotope composition of this species, an opportunistic grazer, and assess whether sex and environmental variables influence such composition.
