Frontiers | Preexposure and Postexposure Prophylaxis of Rabies With Adeno-Associated Virus Expressing Virus-Neutralizing Antibody in Rodent Models

Rabies, a fatal disease in humans and other mammals, is caused by the rabies virus (RABV), and it poses a public health threat in many parts of the world. Once symptoms of rabies appear, the mortality is near 100%. There is currently no effective treatment for rabies. In our study, two human-derived RABV-neutralizing antibodies (RVNA), CR57 and CR4098, were cloned into adeno-associated virus (AAV) vectors, and recombinant AAVs expressing RVNA were evaluated for postexposure prophylaxis after intrathecal injection into RABV-infected rats. At 4 days post-infection with a lethal dose of RABV, 60% of the rats that received an intrathecal injection of AAV-CR57 survived, while 100% of the rats inoculated with AAV-EGFP succumbed to rabies. Overall, these results demonstrate that AAV-encoding RVNA can be utilized as a potential human rabies postexposure prophylaxis. ....

H. pylori strains used, the sequence of 16SrDNA, a ribosomal gene involved in protein production, was analyzed, and the presence or absence of gene mutation was examined. Table 3 shows the number of base substitutions and the number of deletions/insertions in the 16SrDNA sequence. Of these, susceptible strain 3 (S3), amoxicillin-resistant strain 1 (AC1), amoxicillin-resistant strain 2 (AC2), clarithromycin-resistant strain 1 (CH1), and metronidazole-resistant strain 2 (MZ2) were resistant to disulfiram in the growth suppression test. Strains resistant to disulfiram tended to have fewer base deletions/insertions.

Microvessel Density is Increased in Human GC Tissues
To confirm the association between Nrf2/HO-1 and angiogenesis in GC, we also determined the microvessel density of these human GC samples. Consistently, the microvessel density was low in paracancerous tissues, but high in GC. Moreover, the microvessel density was low, moderate, and high in well-, moderately-, and poorly differentiated GC, respectively (Figure 8). Collectively, the Nrf2/HO-1 axis is likely to participate in the malignant process of GC by affecting angiogenesis.

13–15 Calycosin is a fat-soluble component in the dried root of
Astragalus membranaceus, which are organic compounds widely found in nature. Calycosin exerts various anti-tumor activities in vitro and in vivo,
16–18 but its mechanism of action in human GC cells is not clear.
In this study, we evaluated the effects of calycosin on apoptosis, the cell cycle, and migration of GC cells under ROS regulation. In addition, we also determined the roles of MAPK/STAT3/NF-κB pathways in these processes.
Materials and Methods
Cell Lines and Cell Culture
Twelve types of GC cells (AGS, KATO-3, MKN-28, MKN-45, NCI-N87, SNU-5, SNU-216, SNU-484, SNU-668, YCC-1, YCC-6, and YCC-16) and normal human lung cells IMR-90 were purchased from American Type Culture Collection (Manassas, VA, USA), and three normal cell lines (GES-1, L-02, and 293T) were purchased from Saiqi Biotech Co., Ltd (Shanghai, China).
