No safety signals found in pregnant women who received mRNA vaccines
Preliminary findings from a large analysis of pregnant women who received mRNA COVID-19 vaccines showed no obvious safety signals, Centers for Disease Control and Prevention (CDC) researchers reported yesterday in the
New England Journal of Medicine.
Using data voluntarily reported to three US vaccine safety monitoring systems the v-safe after vaccination health checker surveillance system, the v-safe pregnancy registry, and the Vaccine Adverse Event Reporting System (VAERS) the researchers examined outcomes among 35,691 pregnant women who received either the Pfizer/BioNTech or Moderna vaccines from Dec 14, 2020 through Feb 28. Outcomes included local and systemic reactogenicity, participant-reported pregnancy outcomes, neonatal outcomes, and pregnancy- and neonatal-specific adverse events. ....

Scientists tie platelet factor 4 to AstraZeneca COVID vaccine-related clots
UK researchers have uncovered the novel mechanism behind rare abnormal blood clotting seen in some AstraZeneca/Oxford vaccine recipients, according to a study today in the
New England Journal of Medicine.
The study involved clinical and lab evaluation of 23 previously healthy patients who experienced blood clots and thrombocytopenia (low platelet counts) 6 to 24 days after receiving the first dose of the AstraZeneca vaccine. Most clots were cerebral venous thrombosis, while some were arterial thrombosis and venous thromboembolisms such as pulmonary embolisms.
Twenty-one patients had antibodies for clot-promoting platelet factor 4 (PF4) on enzyme-linked immunosorbent assay (ELISA) before heparin administration. They appeared to be of the immunoglobulin G (IgG) subtype. Functional heparin-induced thrombocytopenia (HIT) testing confirmed the positive PF4 ELISA result in five of seven patients ....

Phase 1 and 2 trials show J&J COVID vaccine safe, immune-producing
Interim phase 1-2a results from Johnson & Johnson s COVID-19 vaccine trials show that its vaccine, Ad26.COV2.S, produces a good immune response and safety profile, according to a
New England Journal of Medicine study yesterday.
Unlike the two messenger-RNA vaccines currently in use in the United States, the Johnson & Johnson vaccine uses a recombinant, non-replicating adenovirus 26 (Ad26) with a complete and stabilized spike protein.
A single dose of Ad26.COV2.S elicited a strong humoral response in a majority of vaccine recipients, with the presence of S-binding and neutralizing antibodies in more than 90% of participants, regardless of either age group or vaccine dose, write the researchers. ....
