刘少奇在这3次中原局会议以及其他会议和与领导干部的谈话中，批判了王明的右倾投降错误，联系皖东实际，深入浅出地阐明了中共中央的方针政策。他强调抗日战争要坚持持久战，不能依靠国民党军队，要招兵买马，依靠共产党领导的人民武装；他指出只有建立抗日根据地，才能坚持抗战取得最后胜利；他强调对待国民党制造反共“磨擦”问题……
在刘少奇的领导下，皖东军民积极开展了反“扫荡”、反“磨擦”斗争。1940年3月，刘少奇直接领导了皖东各县抗日民主政府的建立工作，亲自确定了定远县抗日民主政府县长人选。
