第四支队主要活动在津浦路西地区。第四支队经过将军岭战斗、玉屏山袭击战、九龙岗袭击战、张二房出击战等多次战斗，开辟了以定远县藕塘为中心的皖东津浦路西抗日游击根据地。 第五支队主要活动在津浦路东地区，先后两次收复来安城，并取得嘉山老三界等战斗的胜利。经过3个多月的艰苦作战，第五支队开辟了以来安县半塔为中心的津浦路东游击根据地。至此，皖东抗战局面初步打开。 1939年12月初，刘少奇（化名胡服）率中共中央中原局机关抵达定远县藕塘镇附近的新四军江北指挥部。他一方面积极传达和宣传了党的六届六中全会精神，正确解决了皖东乃至华中地区革命斗争方针和策略问题；另一方面，他在第四支队和第五支队等部夺取定远自卫反击战和半塔保卫战的重大胜利后，不失时机地进行了抗日民主政权和根据地的创建工作。
