A C-terminal His-tag was used to express SignalChem’s Recombinant 2019-nCoV Spike protein S1 (Δ69-70, Y453F, D614G) (16-685) in CHO cells. The protein’s purity was found to be approximately 90% by densitometry having a molecular weight of 130 kDa.
The activity has been detected by analyzing the binding potential of the 2019-nCoV Spike protein S1 (Δ69-70, Y453F, D614G) to the immobilized human ACE2 (19-740) protein with the help of a functional ELISA.
Scientific background
Throughout the world, novel coronavirus SARS-CoV-2 has resulted in the pandemic of respiratory diseases (COVID-19). The spike glycoprotein (S) of coronavirus, a type I transmembrane protein that comprises two subunits (S1 and S2) has been known to bind with host cells via the interaction with angiotensin-converting enzyme 2 (ACE2).
SignalChem’s Recombinant 2019-nCoV Spike protein S1 subunit, RBD (Y453F) (319-541), was expressed in CHO cells with a C-terminal His tag. The purity of the S1 subunit was found to be approximately 90% using densitometry and the molecular weight was roughly 39 kDa.
The activity of the S1 subunit has been detected by investigating the binding potential of the 2019-nCoV Spike protein RBD (Y453F) to the immobilized human ACE2 (19-740) protein with the help of a functional ELISA.
Scientific background
The SARS-CoV-2 spike glycoprotein’s receptor binding domain (RBD) identifies the host ACE2 receptor and is a crucial determinant of viral entry and neutralization, while being the most divergent region.

The 2019-nCoV High Sensitivity S1 Protein ELISA Kit v2.0 available from SignalChem is a sandwich enzyme-linked immunosorbent assay that can be used for the quantitative measurement of 2019-nCoV Coronavirus Spike protein.
The S1 Protein ELISA uses a capture anti-S1 protein antibody coated on a 96-well plate. Samples and calibrators are pipetted into the wells, and the target protein contained in the samples and calibrators attaches to the immobilized capture antibody.
The HRP-labeled Detection antibody is added after washing the wells. Once the unbound HRP-antibodies are washed away, a colorimetric substrate solution (TMB) is pipetted into the wells. The absorbance of the color at 450 nm is quantified. The higher the intensity of color development within the wells, the more the amount of target protein bound.
