Walker Fold Road will shut on Monday for around four months.
The work to improve the country road is part of the council's planned borough-wide maintenance programme.
Roadworks involve carriageway reconstruction and drainage works between Chorley Old Road And Edge Lane.
Highway bosses say that Walker Fold Road will be closed throughout the works, signs will be placed on site giving advanced notice of when the closure will be in operation.
The work is expected to take 16 weeks to complete subject to weather conditions.
A spokesman for Bolton Council said: "The council would like to apologise in advance for any inconvenience caused to residents and road users. Residents and businesses will be able to access their properties during the works and have been informed of the plans, although there may be times when access is restricted."
