คณะทันตแพทยศาสตร์ มหาวิทยาลัยมหิดล เดิมมีชื่อว่า คณะทันตแพทยศาสตร์พญาไท พระบาทสมเด็จ พระเจ้า อยู่หัวภูมิพลอดุลยเดช มีพระบรมราชโองการโปรดเกล้าฯ ให้ตราพระราชกฤษฎีกาจัดตั้งคณะฯ ขึ้นเมื่อวันที่ 7 มิถุนายน 2511 เพื่อเพิ่มจำนวนทันตแพทย์ ให้เพียงพอต่อความ ต้องการของประเทศในขณะนั้น คณะทันตแพทยศาสตร์ พญาไท เลขที่ ๖ ถนนโยธี เป็นคณะทันตแพทยศาสตร์คณะที่2 โดยขยายมาจากคณะทันตแพทยศาสตร์คณะแรกที่ถนนอังรีดูนังต์ ต่อมาในวันที่ 2 มีนาคม 2512 มหาวิทยาลัย แพทยศาสตร์ ได้รับพระราชทานนาม"มหิดล" เป็นนามใหม่ของมหาวิทยาลัยตามพระนามาภิไธย ในสมเด็จพระมหิตลาธิเบศรอดุลยเดชวิกรมพระบรมราชชนก เพื่อรำลึกถึงพระมหากรุณาธิคุณที่ทรงมีต่อ การแพทย์แผนปัจจุบันของประเทศไทย.
