1911年（明治44年）、渋沢栄一、伊藤博文ら時の政財界の重鎮の発起により日本初の本格的な洋式劇場として設立。洋の東西を問わない名作舞台が数多く誕生しました。.
大衆性と芸術性を兼ね備えた演劇を目指す開場以来の精神は、1966年（昭和41年）、東宝の演劇担当重役兼、劇作・演出家・プロデューサーの菊田一夫に受け継がれ新装。大規模な舞台機構と重厚な内装は、“東洋一の劇場”と謳われました。「マイ・フェア・レディ」「屋根の上のヴァイオリン弾き」「ラ・マンチャの男」など、世界的な名作ミュージカルが日本で初めて上演されました。現在も「レ・ミゼラブル」「ミス・サイゴン」「エリザベート」「SHOCK」などの大型ミュージカルを中心に、通年にわたりさまざま演劇を上演しています。
