SynNotch-CAR T cells overcome challenges of specificity, heterogeneity, and persistence in treating glioblastoma

Science for just $15 USD. Short circuiting solid tumors Two major hurdles in chimeric antigen receptor (CAR) T cell therapy for solid tumors are ensuring specificity to tumor cells without affecting healthy cells and avoiding tumor escape due to antigen loss. To address these challenges, Hyrenius-Wittsten et al. and Choe et al. developed synthetic notch (synNotch)–CAR T cells targeting solid tumor antigens and used them to treat mouse models of mesothelioma, ovarian cancer, and glioblastoma. In both studies, the authors demonstrated that synNotch-CAR T cells were better at controlling tumors than traditional CAR T cells and did not result in toxicity or damage to healthy tissue. These results suggest that synNotch-CAR T cells may be an effective treatment strategy for solid tumors.

Huntington s Disease: Neural traffic could help understand the disease

E-Mail IMAGE: In Huntington s Disease, axonal transport is defective and this leads to neuronal degeneration. In this work, we found that the protein mutated in Huntington s Disease, called huntingtin (HTT), needs to. view more  Credit: by University of Trento Huntington s disease is a genetic neurodegenerative disorder caused by mutations in the protein huntingtin and characterized by involuntary dance-like movements, severe behavioural changes and cognitive impairment. That neuronal traffic is impaired in this disease has been very well known for several years. But that this deranged trafficking could be ameliorated by increasing huntingtin methylation was not yet known. The findings emerged from an international research work coordinated by the University of Trento and published in

Innovative cancer treatment effective in combating fungal infection in vitro and in mice

Innovative cancer treatment effective in combating fungal infection in vitro and in mice An innovative cell-based treatment for cancer has been found promising for the control of infections caused by fungi. A study published in the journal Cytotherapy reports that the use of CAR (chimeric antigen receptor) T-cells programmed to recognize Cryptococcus spp. fungi was effective in combating infection in vitro and in mice. C. gattii and C. neoformans are present in soil with dead organic matter and places contaminated by the droppings of pigeons and other birds. They cause systemic mycoses in the human organism. They can infect the lungs and central nervous system, causing meningitis or meningoencephalitis. The symptoms vary according to the site of the infection, which can be fatal. Transmission occurs by inhalation of the fungi.
